Can someone explain in short by giving examples as in

building a town with or without builders?
How does a builder or landlord help?
If a town has been detroyed and I place a builder will it rebuild faster?

Builder has use only when you need to build fast some building. Generaly no use at all

Landlord has mostly good use at the start of game when you need more food very fast but then he's almost useless as you have enough of both food and people.

Builder improves rapidity of building buildings and restoration of plundered villages
Landlord raises food production as well as population growth and in higher levels also number of people. In unmodded game almost useless.

But in the Holy Rome mod 1.8.1 I finally made them useful: you lack food, population in provinces is much lower so you need to improve it. And when your villages in province are plundered, you don't produce food, but consume it. And when the province goes to negative food production and there is no food stored, you can't even refill your squads - so using builder to restore the villages as fast as possible is useful.

But without this modifications there's no need to use them at all.

Thanks a lot. I am still trying to figure out the game. I believe if this game had the "zoom" feature that most of us have been ranting about it would be the best game ever made on this subject. Heck just add the zoom feature and came out with KOH-2

I find both roles very useful, but usually it's a late game strategy.

I only use Royal Family for these roles, preferably the King or first Prince.
It's usually just a temporary role, so you want your Landlord/Builder to be as effective as possible. You should be churning out a ton of books by the latter stages of a game, so upgrade them to 5-stars as soon as possible.
Landlord is useful when you need population, and fast. You have (or need) crop, and have all the prerequisites for troops, but you just don't have the population. For example, you've been heavily recruiting from one province, and you are very low on crop and peasants to recruit from. Throw a Landlord on the province, and watch your crop and peasant production skyrocket. ... or if you know some one is coming to attack one of your castles, but you are low on troops and crop, and Landlord will help you restock quickly and avoid losing the siege.
Landlords are also useful when you are churning out a lot of cavalry. They will refill your food stocks very quickly.
Builders are useful when you inherit a scorched-earth province and had to rehabilitate it ASAP. A 5-star builder is the best help for that.
A Builder will also help speed up construction, but in the latter stages of a game, you should have lots of gold.

Generally, Landlords are useful in crop-poor provinces, and Builders are useful in worker-poor provinces.
